Design Notes
While creating this construct, there was a mystery sequence that appeared bewtween the Spe1 and Pst1 sites. If you cut with Spe1 and Pst1, this shouldn't be a problem. This is just a disclaimer for sequencing. Here is the sequence from the First Spe1 site through the Pst1 site.
ACTAGTAGCGGCCGCTACTAGTTCAGACCACGCTGATGCCCCCTGCAG
There is a mystery 27 base pairs between them, including an additional Spe1 site.
